果蝇是从细胞到组织水平研究形态学分子机制的重要模式生物。翅是果蝇组织中最大的器官,具有表型明显、方便观察等优点,因此果蝇的翅成为研究信号转导,器官形成机制的首选材料。果蝇属完全变态(holometabolous)昆虫,其成虫的翅是由幼虫时期的翅成虫盘(wing imaginal disc,又称翅原基)发育、分化形成。昆虫翅的形成利于昆虫的迁徙、繁殖、转移,扩大生活范围。因此,研究昆虫翅发育不仅有助于我们了解昆虫变态发育过程,而且还有益于我们深入探究器官形态发生的分子机制和生物进化发育提供了广泛的理论依据。果蝇anchor(CG7510)在哺乳动物的同源物是GPR155,是一种孤儿G蛋白偶联受体。GPR155与亨廷顿病(Huntington disease)和自闭症谱系病(autism spectrum disorders,ASD)发病机制有关。另外,研究表明anchor敲除的果蝇翅发育异常,但是对翅脉的形成及翅发育的调控机制没有任何报道。Jumeau(Jumu)是Forkhead(Fkh)转录因子家族成员之一,该蛋白参与果蝇心脏发育,维持造血和免疫系统稳态等。研究表明,jumu低表达引起果蝇复眼、翅与刚毛的发育异常,但是该基因在果蝇翅发育中功能与机制还没有报道。本文主要以anchor和jumu突变体或转基因果蝇为实验材料,通过分析成虫翅、Anchor与Jumu在果蝇翅原基和蛹期翅组织中的定位,并采用免疫组织染色的方法对幼虫时期翅原基的细胞增殖与凋亡、蛹期翅脉间区细胞、翅脉与刚毛发育,以及上位遗传等方面进行了分析。本研究主要获得以下结果:1、anchor低表达引起果蝇翅面积增加、翅脉发育异常。2、原位杂交结果表明,anchor基因广泛表达在幼虫翅原基与蛹期翅组织中。3、anchor低表达引起幼虫翅原基细胞分化明显增加,且Wg的表达量升高。4、anchor低表达时,幼虫翅原基的BMP信号异常激活,其中p-Mad、omb、sal的表达量明显升高,但Dad与brk的表达量没有变化;蛹期翅的p-Mad和Dad的表达量升高,且brk的表达量降低,这说明蛹期anchor对BMP信号的影响更显著。5、上位效应分析表明anchor位于Dpp配体平行或其上游调节BMP信号途径。6、jumu突变体成虫翅面积减小、翅缘缺失以及翅面刚毛增加。7、抗体染色表明,Jumu定位于幼虫翅原基WM(wing margin)附近和蛹期脉间区的细胞核内。8、jumu突变体翅原基细胞异常增殖、Wg水平升高,并产生大量凋亡细胞;并且,p-JNK与puc的表达量升高,引起JNK信号异常激活;但是成虫翅缘缺失与JNK信号诱导的细胞凋亡无关,而是与Cut表达量下降引起的Wg信号应答机制有关。9、jumu突变体蛹期F-actin表达量升高,DE-cad与Rho1的表达量降低从而导致刚毛增加。以上结果说明,anchor与jumu基因均参与到翅发育的过程中,对翅的形态发生与细胞分化起到关键性作用,为进一步调查GPCRs在器官发育中的功能提供了新线索,同时也为深入研究Fox家族成员在器官发育形成中发挥的功能提供理论基础。
