已知“体温调节”(thermoregulation)对机体生存起到关键作用。温度敏感型TRP通道(thermosensitive TRPs,thermo-TRPs)是一类与温度感知密切相关的蛋白质分子,动物随着环境温度的不同,能主动调节thermo-TRPs的表达量,于是让机体在环境中保有更高的存活机率。根据各个thermo-TRPs蛋白质感知温度的阈值,它们被区分为3类:1)热敏TRP通道蛋白质(the heat-activated TRPs,h-TRPs),例如TRPV1、TRPV2、TRPV3、TRPV4;2)温热敏TRP通道蛋白质(the warm-activated TRPs,w-TRPs),例如TRPM2、TRPM4、TRPM5;3)冷敏TRP通道蛋白质(the cold-activated TRPs,c-TRPs),例如TRPA1、TRPC5、TRPM8。“冬眠”是动物用于对抗饥寒交迫并且经过长时间演化出的生存本领。在近期研究中我们发现h-TRPs在蝙蝠冬季蛰伏时表达量上升,而w-TRPs和cTRPs在蝙蝠夏季活跃时表达量上升;由此可见,处在低温环境下,冬眠中的蝙蝠可能对感知温度升高更为灵敏;而处在夏季温度高的环境下,蝙蝠对感知温度下降可能更为灵敏;研究结果同时提示thermo-TRPs在哺乳动物冬眠中扮演重要角色。为了深入研究thermo-TRPs在大足鼠耳蝠(Myotis ricketti)冬眠与非冬眠时期的表达与调控模式,我们从转录因子(transcription factors,TFs)调控基因表达的方向着手。在本论文中,我们用生物信息学方法推测出某些POU和HOX家族转录因子可能调控thermo-TRPs基因的表达,再通过实时荧光定量PCR反应(quantitative real-time fluorescence PCR,qPCR)、蛋白质免疫印迹法(Western Blotting,WB)、双荧光素酶报告基因实验和小鼠实验探究之后,我们首次1)呈现了蝙蝠在不同生理状态下,其thermo-TRPs和相关TFs的表达模式;2)证实了POU5F1可以调控TRPC5基因的表达;3)发现了HOXC13与TRPV2、TRPV3、TRPV4的表达有正相关;4)推测小鼠中的Trpa1很可能是热敏感通道。本篇研究首度揭示了冬眠蝙蝠体温调节的重要分子机制。
