Wheat(Triticum aestivum) is one of the main food crops in China.Drought stress can destroy the balance of water potential and lead to osmotic stress in wheat.Plants can sense the signal of osmotic stress and then regulate the gene expression to protect the plant growth.In this study,a total of 160 differentially expressed genes related to osmotic adjustment under PEG induced stress were screened through transcriptome sequencing in the drought resistant cultivar Qingmai No.6 of wheat.Among these genes,91 and 69 genes were up-and down-regulated,respectively.Two drought resistant candidate genes,TaAGXT2 and TaGS1-3 showed high homology with Aegilops tauschii,were identified for further study.The full length of these two genes was obtained by RACE technology.And their tissue expression patterns were detected using qRT-PCR.The result showed that the abundances of TaAGXT2 was decreased in leaves and increased in roots under drought stress,whereas the expression of TaGSl-3 was just the opposite.In addition,the gene’s structure,conserved domain and phylogenetic relationship were analyzed by bioinformatic analysis.Furthermore,their functions were verified by overexpression and RNAi technologies.TaAGXT2 overexpression plants showed higher photosynthetic level than the wild plants,while the RNAi plants had lower photosynthesis than the wild plants.TaGS1-3 overexpression plants manifested enhanced N assimilation efficiency compared with the wild type.As a result,the function of TaAGXT2 is involved in regulating photosynthesis,and TaGS1-3 plays an important role in the N assimilation in wheat.The current results provide important genes for drought resistance improvement in wheat.
