The 2C Ser/Thr phosphatases regulate development, aflatoxin biosynthesis and pathogenicity in Aspergillus flavus

Protein phosphorylation and dephosphorylation excuted by kinases and phosphatases, respecitively, is known to play important roles in the regulation of various cellular processes in eukaryotes. However, compared with kinases, little has been known about the function of phosphatases in filamentous fungi. Type 2 C Ser/Thr phosphatases(PP2 Cs) is consider to be a main subfamily of protein phosphatases. Here, 7 members of PP2 C phosphatases were identified and analyzed in Aspergillus flavus, a fungus that produces one of the most carcinogenic secondary metabolites, aflatoxin B1(AFB1). Firstly, a inhibitor(Sanginarine chloride) of PP2 C phosphatases was used to treat with A. flavus, and the result showed that Sanginarine chloride could affect its development and aflatoxin biosynthesis. Then, all PP2 C phosphatases single gene deletion mutants were generated using homologous recombination strategy. The ptc1 and ptc2 deletion mutants exhibited dramatically reduction in conidiation, aflatoxin production and pathogenicity, while an enhanced production of the sclerotial was observed in Δptc1 mutant. Additionly, double knockout mutant(Δptc1/Δptc2) displayed more significant defects in conidiation, sclerotial, aflatoxin production and pathogenicity than that of single deletion mutants,which were consistent with the expression levels of related genes. However, no recognizable changes were obtained in other PP2 C phosphatase mutants, when compared with WT strain. Overall, our results indicated that PP2 C phosphatases ptc1 and ptc2 could play an important role in development, secondary metabolism and pathogenicity in A. flavus, which could be displayed as potential targets for preventing this fungal pathogen.
