Gene clone and eukaryotic expression of a putative effector g10687 from Cylindrocladium parasiticum

The objective of this study is to clone and construct eukaryotic expression plasmids of a putative effector g10687 from Cylindrocladium parasiticum（isolate: Cylpar14017）. The full length DNA and c DNA of g10687 were cloned by PCR. Bioinformatics analysis revealed that the gene contained 812 bp, of which the ORF was 753 bp encoding 251 amino acids with hypothetical molecular weight of 26 kDa.The deduced amino acids sequence of the protein had a transmembrane region and signal peptide. The complete ORF of g10687 was amplified by RT-PCR, and ligated to the eukaryotic expression vector pGAPZαA. The recombinant plasmid was identified successfully by restriction enzyme digestion and sequencing. Subsequently, the recombinant plasmid was transferred into Pichia pastoris strains of X33 by Gene Pulser Xcell. According to SDS-PAGE analysis, target protein of the recombinant g10687 protein could be observed after 24 h hours induced by Glycerol. Based on the result, it should be possible and potential to express this effector of g10687 by Pichia pastoris.
