产肠毒素大肠杆菌3×STa-ovalbumin融合蛋白的血清学评价

论文摘要

STa-ovalbumin化学偶联物是目前ELISA方法检测STa抗体唯一应用的包被抗原,但是它的制备复杂并且效率低。研究以3×STa-ovalbumin融合蛋白为包被抗原,检测STa抗体阳性血清,探讨该融合抗原的最佳包被浓度和对STa抗体检测的敏感性和特异性。Western blot分析结果显示,该重组蛋白可被STa阳性血清特异性识别。抗原包被剂量优化试验表明,每孔25 ng的包被剂量为最佳包被剂量。敏感性和特异性试验表明,选用该融合蛋白作为包被抗原ELISA检测STa抗体具有与STa-ovalbumin化学偶联物相同的效果。研究结果表明,3×STa-ovalbumin融合蛋白可以作为STa抗体ELISA检测的替代抗原,也将促进STa抗体检测的标准化和产肠毒素性大肠杆菌疫苗的研发。
