Phosphorylation of Suppressor of fused(Sufu) is modulated by protein phosphatase 4 regulatory subunit 2(Ppp4r2) in Sonic Hedgehog signaling

Protein reversible phosphorylation is one major regulator of life activities. Sufu, a major downstream regulator of Hh signaling, is stabilized under dual phosphorylation of PKA and GSK3β. Its phosphorylation is reduced with the activation of Shh signaling. However, the phosphatase in this reversible phosphorylation has not been found. Through a proteomic approach, we identified Protein phosphatase 4 regulatory subunit 2(Ppp4r2) interacted with Sufu. Shh signal promoted their interaction in the nucleus. Protein phosphatase 4(PP4) dephosphorylates Sufu, thereby promoting its degradation and downstream Gli1 transcriptional activity. Finally, PP4-mediated dephosphorylation of Sufu promotes cell proliferation of medulloblastoma cells. Expression of PP4 is significantly increased in the SHH-subtype medulloblastoma and positively correlated with Shh pathway target genes. In summary, our findings reveal PP4 modulates Sufu phosphorylation in Shh signaling.
